Effect of RAS Polymorphism on Essential Blood Pressure Pharmacogenetic Association of RAS Polymorphism on Essential Blood Pressure in Relation to Enalapril/Lisinopril among Malay male newly diagnosed hypertensives Abstract Objective: It has been suggested that genetic backgrounds, which have an association with essential hypertension, may also determine the responsiveness to ACE inhibitor. We determined the association of angiotensin-converting enzyme (I/D, G2350A), angiotensinogen (M235T, T174M and A-6G) and renin (Bg/I and Mbo/I) gene polymorphisms with essential hypertension and the relationship between genetic variant of interest and high blood pressure response to ACE inhibitor (enalapril, lisinopril) in patients with essential hypertension subjects from Seremban, Malaysia. Methods: A newly hypertensive Malay male population (n=142) was recruited for a mono-trophy[ah1] pharmacogenetic study. Hypertensive patients were treated with ACEI drugs, particularly enalapril or lisinopril alone. We differentiated between those who controlled their HT with those who did not. Each group’s characteristics were compared to determine the risk of non-controlled HT associated with RAS polymorphisms by adjusting for different variables. Results: Statistically significant associations of I, G, T and M alleles were observed with essential hypertension in I/D, G2350A, M235T, and T175M. The decrease in systolic blood pressure and diastolic blood pressure after 24 weeks of treatment of the patients carrying II, GG, and TT genotypes was greater than the groups carrying DD, AA, MM and MM genotypes. In contrast, no significant difference was shown between renin gene polymorphisms (Bg/I and MboI). Conclusions: Although this study shows a possible association of polymorphisms of RAAS genes[ah2] with the risk of non-controlled HT in ACEI-treated patients and indicates the importance of all components in this system in regulating HT, it needs to be replicated in other data sources. In Malaysia, the National Health and Morbidity Survey (NHMS) 2013 has shown that the prevalence of hypertension in adults ≠¥18 years increased from 33.2% in 2006 to 35.7% in 2013 [2]. Furthermore, the prevalence increased from 42.6% to 43.5% for those >30 years old. Unfortunately, 60.6% of total hypertensives were â€Å"undiagnosed† [3]. These poor rates of high blood pressure (BP) control are not explained by the lack of treatment, as one study estimated approximately 30% of treated hypertensive patients take one antihypertensive drug, 40% take two antihypertensive drugs and 30% take three or more antihypertensive drugs [4]. These data suggest that the present trial and error approach for high blood pressure management is suboptimal, and alternative approaches for identifying the optimal antihypertensive regimen in a specific patient are needed. Using genetic make-up of an indiv idual along with the association between single-nucleotide polymorphisms (SNPs) and angiotensin-converting enzyme (ACE) inhibitors for hypertensive response offers a new preventive approach to lower adverse drug interaction risk. A renin-angiotensin system (RAS) is an important component of blood pressure regulation, and it has been suspected to be involved in hypertension [5]. Moreover, the major active peptide of the RAS is angiotensin II. Produced from the precursor molecule, angiotensinogen (AGT), via an enzyme cascade involving ACE enzyme, angiotensin II exerts numerous effects on the homeostatic regulation of blood pressure, the vast majority of which are mediated via the angiotensin II type 1 receptor (AT1R) [6]. The presence of polymorphisms in the ACE, AGT and renin (REN) genes of the RAS has been associated with adverse EH changes in several studies [7,8,9]. For example, the insertion/deletion (I/D) polymorphisms of the ACE gene have been associated with increased blood pressure, urinary albumin excretion (UAE) and target-organ damage in hypertensive patients [10]. Moreover, ACE G2350A gene polymorphisms in exon 17 were reported as a remarkable genetic variant mostly associated through hypertension with an average increase of 3.2 mmHg in SBP by having the G allele [11]. It has been reported that the presence of A-6G polymorphism of AGT gene among Chinese hypertensives increased body weight gain in hypertensive patients [12]. The Mb/I and Bg/I polymorphisms of the REN gene have been associated with hypertension, left ventricular hypertrophy, aortic stiffness and exaggerated vasoconstriction; additionally, the Bg/I polymorphism in the same gene appears to confer protection against the development of microalbuminuria in patients with hypertension [13]. The purpose of the present study was to evaluate the impact of four RAS gene polymorphisms on the antihypertensive response in newly detected hypertensives receiving two ACEIs (enalapril and lisinopril). The polymorphisms investigated were A-6G, the A for G substitution of the AGT gene 6 nucleotides upstream from the start site; the ACE I/D polymorphism corresponding to an insertion or deletion of a 287bp alu repeat; and two polymorphisms of the REN gene, Bg/I and MboI and both in the coding area of intron 9. There are compelling reasons hypothesizing that variations in genes of the system may be predictive of variations in BP response. Therefore, genetic variation of the RAS has been investigated in relation to antihypertensive response to ACE inhibitors in various population and dosage (Table 1) as the most common lowering BP agent in Malaysia [14]; however, previously publications have had somewhat conflicting results elsewhere [15-19]. We hypothesized those genetic polymorphisms in RAS genes, including ACE I/D, G2350A, AGT M235T, T174M, A-6G along with REN MboI and Bg/I were associated with the incidence of EHT. Therefore, the aim of this pharmacogenetic study was to investigate the association between seven RAS gene polymorphisms of interest among 142 newly diagnosed Malay male hypertensives that never took BP medications. They were treated once daily for 24 weeks with 20 mg of enalapril or lisinopril. MATERIAL AND METHODS Patient Populations Malay male patients >18 years of age with three-generation Malay family who were newly diagnosed with essential mild-to-moderate hypertension were collected from clinics for non-communicable disease Seremban, Malaysia. The information includes age (25-60 years old), onset (25-60 years old), systolic BP > 140 mmHg and a diastolic BP > 90 mmHg on 2 consecutive visits for those untreated, absence of secondary forms of hypertension. Subjects with a history of diabetes mellitus, renal failure and major infectious disease were excluded. They had no metabolic or endocrine disorder, as well as any acute illness. They were not on any antihypertensive treatment and were drug-naive patients. Written informed consent was obtained from each patient before being included in the clinical trial, and patient’s identity was kept strictly confidential. A specific consent form was requested for genetic testing permission. BLOOD PRESSURE REDUCTION PATTERN Lifestyle Modification For patients, lifestyle modification for a period of three months was advised. The patients were seen three times during this period to assess the efficacy of the non-pharmacological management including weight loss, regular exercise, and ingestion of a high-fiber, low-fat, and low-salt diet. Follow-up Mon-trophy[ah3] Management Dispensed ACEIs (lisinopril or enalapril) on the same date for individuals have recorded. Each patient received lisinopril or enalapril (20 mg, once daily) for 24 weeks on a regular basis. Patients’ BP was measured using the same device and protocol; follow-up visits were made 12 times (once per two weeks). Of the 152 patients, 10 lost to the follow-up along monitoring due to relocation, travelling and/or change of medication. Eventually 142 hypertensives (92.6%) completed the study. This subsample was divided into two groups; individuals whose HT was not controlled as the non-responded (n=35), and individuals whose HT was under control as the responded (n=107). Genotyping procedures A blood sample was taken in two separate tubes; one was used for colorimetric analysis of total cholesterol (TC), high density lipid profile (HDL), low density lipid profile (LDL), triacylglycerol (TG) and fasting glucose (FBG) using Diays Commercial Kits (Diagnostic System, GmBH, D66559 Holzheim, Germany), and the other test tube contained venous blood samples collected on EDTA was subjected to DNA extraction, which was obtained from individuals in the morning after a minimum of 8 hours fasting at the time of randomization. Eventually, the samples were stored at -20 °C for further molecular and biochemical analysis. DNAs were extracted from 5 mL blood samples as explained elsewhere [20]. ACE I/D polymorphism was genotyped using DNA amplification with oligonucleotides as described elsewhere [21]. For ACE G2350A, DNA amplification followed the approach by Zhu et al. [22]. Reactions were conducted using DNA amplification in a final volume of 25 mL containing 20 pmol of each primer, 0.4 mmol/L of each deoxynucleotide triphosphate (dNTP), 2 mmol/L of MgCl2, 1XTaq buffer and one unit of NEB Taq DNA polymerase (New England Biolabs, Beverly, MA, USA). The PCR cycling conditions were carried out in an iCycler machine (BioRad Laboratories, Hercules, CA, USA). Were chosen[ah4] for genotyping using the polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) approach, and the details are presented in Table 2. Eventually, DNA fragments were stained in ethidium bromide and visualized by Alpha Imager (Alpha Innotech, San Leandro, CA, USA) under ultraviolet (UV) light. In order to build a good team there needs to be other elements besides leadership in order to carry out their plans: such as tools and technology, diversity, and the proper personalities to keep the team intact. Everyone’s personality is different, and that in turn leads to the roles they play within the team. The diversity of the different personalities and the diversity of the departments they work in can lead to good and bad connotations. As a team everyone should have shared leadership. The tools are needed to help the team create, develop, and execute the plans they come up with. In order to create plans of action, the team needs a harmonious work environment and it should be in a place that no one has connections to, and that everyone feels comfortable in. They should be well stocked with all the office supplies all of them will need. Even though we think of office supplies as pens and pads of paper, office supplies does not mean that in every company. If they are working in a car company, one of the office supplies they will need is large amounts of clay in order to come up with new models for the upcoming year. If they are working for Blue Bell, they might need flavorings and candy. All of the supplies should be readily available because when they get on a roll, the last thing they want to do is stop and go searching for something they need. Computers are a very important part of a team. They can connect all of the team members who may not all be in the same off ices. An example of this would be video Team A 2 conferencing. An international company might create a team of individuals who live and work all over the world, so in order to get them all together they would have to video conference. Another helpful function of the computer is the internet. The internet supplies us with endless amounts of data, and is a useful tool to help with research on any subject. Another tool that is quite important and ties into computer and the internet is software. One of the new plans the team might come up with to execute to the whole company is a new software program. As a result, this understanding acknowledgment and respect of differences bonded our partnership. When a variety of people come together to solve problems the forum becomes rich and robust. In conclusion, by having the proper and an adequate supply of resources and personalities that offer unique traits, and members that are diverse, teams are most effective in reaching their goals. By no means is teamwork easy, it requires its members to have and keep an open mind, to have tremendous patience, to take ownership and responsibility, to ask questions, to take criticism, and to consider ideas that are not your own, all the while keeping a clear definition of the goal. Felsic rocks with higher silica content melt at lower temperatures than mafic rocks which contain less silica and more iron/magnesium.Lower the confining pressure: a reduction in pressure can induce a hot rock to melt. A reduction in confining pressure on the h ot upper mantle is thought to generate the basaltic magmas which intrude into the oceanic ridge system to form ocean crust; refer to Figure 4. 15. Add water: the presence of water in a rock can lower its melting temperatures up to a few hundred degrees. Water released from rocks subducting into the mantle along convergent plate boundaries is thought to be an important factor in magma generation at convergent plate boundaries.As subduction begins water trapped in the rock is subjected to increasing temperature and pressure. Sedimentary basins result from rifting, thermal sag, and flexure of the lithosphere. The sedimentary stages of the rock cycle involve the overlapping processes of weathering, erosion, transportation, deposition, burial, and diagenesis. Weathering and erosion produce the clastic particles and dissolved ions that compose sediment. Water, wind, and ice transport the sediment downhill to where it is deposited.Burial and diagenesis harden sediments into sedimentary rocks via pressure, heat, and chemical reactions. The two major types of sediments are clastic and chemical/biochemical. Clastic sediments are formed from rock particles and mineral fragments. Chemical and biochemical sediments originate from the ions dissolved in wate r. Chemical and biochemical reactions precipitate these dissolved ions from solution. Understanding the characteristics of sediments and modern sedimentary environments provides a basis for reconstructing past environmental conditions using the rock record.Sedimentary structures like bedding, ripple marks, and mud cracks, provide important clues about the sedimentary environment. Diagenesis transforms sediment into sedimentary rock. Burial promotes this transformation by subjecting sediments to increasing heat and pressure. Cementation is especially important in the lithification of clastic sediments. The classification of clastic sediments and sedimentary rocks is based primarily on the size of the grains within the rock. Assessment is a systematic process of collecting information or evidence about a student’s progress towards meeting the learning expectations. Assessment is embedded in the instructional activities throughout a unit. The expectations for the assessment tasks are clearly articulated and the learning activity is planned to make that demonstration possible. This process of beginning with the end in mind helps to keep focus on the expectations of the course. The purpose of assessment is to gather the data or evidence and to provide meaningful feedback to the student about how to improve or sustain the performance in the course. Scaled criteria designed as rubrics are often used to help the student to recognize their level of achievement and to provide guidance on how to achieve the next level. Although assessment information can be gathered from a number of sources (the student himself, the student’s course mates, the teacher), evaluation is the responsibility of only the teacher. For evaluation is the process of making a judgment about the assessment information and determining the percentage grade or level. Assessment is embedded within the instructional process throughout each unit rather than being an isolated event at the end. Often, the learning and assessment tasks are the same, with formative assessment provided throughout the unit. In every case, the desired demonstration of learning is articulated clearly and the learning activity is planned to make that demonstration possible. This process of beginning with the end in mind helps to keep focus on the expectations of the course as stated in the course guideline. The evaluations are expressed as a percentage based upon the levels of achievement.

The only colors notable is the brown used to darken the prolific amount of nails in the mouth of our main attraction, and the red painted in horrific amounts, ejecting from any open space that the lips could endure. * This painting in particular, I could not completely ingest, and with only scant knowledge of her country (other than the stolen and self-proclaimed Presidency, by once attorney turned Prime Minister; Fidel Castro), I did not wish to assume. However, upon researching Bruguera further, I confirmed her rebellion, and alas; was able to enjoy her diverse techniques, yet insufficiently represented; fight for her country. 6) The example for my 6th piece of art, I would instead, like to use one that I would personally have to top the list with; a portrait of my son, my baby boy; Robert Emiliano Lara, so named after my exceptionally unique Uncle Robert and also after Emiliano Zapata, by his father. Zapata is, in fact, the owner of one of our favorite quotes: I would rather die standing, than to live on my knees. * Canvas generously donated by: Jose N. Lara, outstanding husband and exceptional father, a cut exceeding the rest! * Last of all, but of the utmost significance; my association of the aforementioned art, to Oscar Wlde, the mischievous, yet well-to-do, witted writer. * Again, going beyond what I have in print before me and unable to control my curiosity, I research more of Wildeâ„ ¢s quotes, of which I will share: * All that I desire to point out is the general principle that life imitates art far more than art imitates life. * Every portrait that is painted with feeling is a portrait of the artist, not of the sitter. * Illusion is the first of all pleasures. * Morality, like art, means drawing a line someplace. * Art is the most intense mode of individualism that the world has known. * What I see / My take: Considering that he had a torturous life, hidden from his true self for the liking of others, ultimately dying when finally free, I believe he may have agreed that; life is never completely understood, reminiscent of my confusion in my interpretation and understanding of what the art was truly representing. Were they emulating their lives, or were their lives emulating their art Just as Wilde expressed, the portrait is for the artist and not the sitter. Also, in stating that illusion is the first of all pleasures, was he then stating that he portrayed himself as something contradictory to the truth I despondently, would have to agree this to be true. In his time, the late 1800â„ ¢s, he was shamed by society, accused of living immorally, and finally imprisoned simply for being himself, one who enjoyed relations with other men. His quote concerning morality, I am sure was written in an unsettled frame of mind, but it is his quote relating individualism to art, that I believe unveils his true character; I think that it was only through his art that he could, in the end; be himself; an individual without a label.

We've marked four policies towards the Subject Tests: required, recommended, considered, and alternative. Required means you have to take the SAT Subject Tests. In a few cases, as you'll see, colleges will accept the ACT in lieu of the Subject Tests. Some colleges require one subject test, while other colleges (typically those that are more selective) require two. Recommended is a little vague, as it leaves the decision up to you whether or not to send SAT Subject Test scores. If you can achieve high scores or want to make up for a low SAT score, then it's a good idea to send Subject Test scores to these schools. The company consistently retains the merit of being the number one regularly scheduled program on cable TV with Raw on Monday nights on TNN, and Smackdown! Is the highest rated show on UPN. It airs on Thursdays. Revenues consist principally of attendance at live events, sale of television advertising time and sponsorships; domestic and international television rights fees, pay-per-view purchases and direct sale of merchandise. Stock for WWE can be purchased through any brokerage or online investing service. Minimums and fees will vary. Investors Choice Plan enables stockholders to purchase shares and reinvest the cash dividends paid on shares. For the past nine weeks stock prices for World Wrestling Entertainment, INC have fluctuated between a high of $14.40 at the end of April to a low of $11.52 towards the end of May. For the month of June stock prices fluctuated between $12.37 for a low and $13.45 for a high. Although stock prices are not as stable as the corporation had hoped, it expects operating profits to more than double from the prior year. WWE anticipates revenue of $375 million, compared with $374 million in the prior year. It expects operating income of $73 million, up from $27 million a year earlier. Net income should range between $44 million and $48 million, or 64 cents to 70 cents a share, compared with a net loss of $19 million, or 28 cents a share, a year earlier. I believe the corporation and management team of World Wrestling Entertainment, INC has done a lot for its fans. Not only does WWE provide live and televised entertainment, they also get involved in matters that are important in the lives of their fans.

Ebonics INTRODUCTION The main topic of this paper is the USA, and I have chosen to concentrate on a fairly new issue, the language know as Ebonics. There have always been changes in the English language. This is how the language came about and evolved from standard British English to American English. During the last few years, as the world has become more sensitive to the rights of minorities, women, animals, etc. a new form of changes has taken place. These changes have become known as Political Correctness. Ebonics is the political correct version of Afro-American English. I intend to show that – and comment on how – racialism and competition affects a society and how this, in the case of Ebonics, is actually happening in today's America. Nuuk February 1997 Ral Fleischer THE QUESTIONS ASKED I will attempt to answer the following questions about Ebonics; - what is Ebonics? - what are the underlying reasons behind Ebonics? - who is prospering from Ebonics? What is Ebonics? Most people outside of America have at most but a vague idea of what Ebonics is all about. Apart from being a buzzword in American media since December 1996 what are the fundamental concepts behind this expression? Where, how and when did it start, and who started it? What are the underlying reasons behind Ebonics? To fully understand Ebonics, some historical background is needed. One has to have some knowledge on how the English language has developed in America. Furthermore one has to be aware how the American society is loaded down with the influence of stereotypical thinking, racialism and competition. In his autobiography, the famous black spokesman, Minister Malcolm X, portrays the undisguised hostility that exists between white and black people in America. He illustrates this conflict with the following words: "†¦You cannot find one black man, I do not care who he is, who has not been personally damaged in some way by the devilish acts of the †¦ white man!. The greatest miracle †¦ in America is that the black †¦ has not grown violent †¦ they would have been justified by all moral criteria, and even by the democratic tradition" (The Autobiography of Malcolm X, page 371 & 349) Is Ebonics simply the democratic consequence, a black non-violent upraise against the "devilish acts of the white man" as as foreseen by Malcolm X? Oakland Unified School District is providing its teachers and parents with the tools to address the diverse languages the children bring into the classroom. 3). The District's objective is to build on the language skills that African-American students bring to the classroom without devaluing students and their diversity. We have directly connected English language proficiency to student achievement. 4). The term "genetically-based" is synonymous with genesis. In the clause, "African Language Systems are genetically based and not a dialect of English," the term "genetically based" is used according to the standard dictionary definition of "has origins in."
